Rubatex Pipe                     Since there are several variables involved in the coating process, it was of interest to optimize these conditions. The core tablets were prepared by direct compression method and was found to possess good characteristics. This study used two factor and three level design to allow a small number of experiments required for optimization. The coating was conducted in both the perforated pan coater and the fluidized bed bottom spray coater. For each type of equipment, the effects of two factors, spray rate (g/min) and inlet temperature (°C), were investigated. The optimum condition was performed with two and three dimensional plots. Percent loss of coating solution and film adhesive force were the criteria for the designation of optimum condition. The adhesive force increased with the increased inlet temperature and the decreased spray rate, and at the value of not less than 7 kg the tablets were non-friable. This value was assigned to be one criterion. To obtain the lowest percent loss of coating solution, the pan coater should have the spray rate of 11.68 to 14.24 g/min and inlet temperature of 99 to 103°C and the bottom spray coater should have the spray rate of 13.6 to 15.25 g/min and inlet temperature of 45 to 49° C.                     Effect of process variables on hydrochlorothiazide granules and tablets were investigated in this study. Three granulators, i.e., sigma blade mixer, planetary mixer, and fluidized-bed granulator were used. A 140 ml portion of 10% w/w aqueous solution of povidone K-30 was added to powder bed as binding agent. One of the process variables was added amount of water to produce acceptable granulations. The other process variables were mixing time, agitation intensity, product temperature and spray rate depending on types of granulators. Three dimensional plots of process variables against granule and tablet properties were generated to demonstrate their relationship. Amount of water added was found to be the major factor which regulated properties of granules produced in sigma blade mixer and planetary mixer and their corresponding tablets. Both spray rate and product temperature exerted predominant effects on the granules and tablets. Tablet weight variation, friability, and dissolution were employed as criteria to determine the optimum conditions for each granulator. Optimization technique was found to be a useful rool to define each individual process variable suited for the granulator used.
